InnovaPrep® Elution Fluids are formulated for use with Wet Foam Elution™ technology to recover collected or retained material into a small-volume liquid sample for downstream analysis.
Because the selected Elution Fluid becomes part of the recovered eluate, formulation choice is more than a recovery step. The fluid should support efficient recovery while also fitting the biological requirements of the target and the analytical method that follows.
The most useful question is not simply, “Which Elution Fluid gives the best recovery?”
Which formulation best supports the complete workflow—from recovery through downstream analysis?

What Is an Elution Fluid?
An Elution Fluid is the recovery fluid used during Wet Foam Elution™.
InnovaPrep Elution Fluids are supplied in pressurized canisters designed for compatible AirPrep™ and FluidPrep™ recovery systems. During Wet Foam Elution, Elution Fluid and gas generate a controlled foam that contacts the collection or concentration surface and transfers retained material into the recovered liquid sample.
Compatible recovery formats include:
- AirPrep™ collection media
- CP Select™ Concentration Tips
- EasyElute™ LVC collection cartridges
- Other compatible Wet Foam Elution collection or concentration devices
The recovered eluate therefore contains both the recovered target-bearing material and the selected Elution Fluid.

Why Elution Fluid Selection Matters
Collection and recovery are linked but distinct parts of the workflow.
The collection or concentration device determines what material is captured or retained. Wet Foam Elution then transfers retained material into the liquid sample used for downstream analysis.
The selected Elution Fluid can therefore matter for more than recovery alone. Depending on the workflow, formulation choice may affect:
- target recovery
- biological integrity
- viability or infectivity
- compatibility with nucleic acid extraction
- compatibility with culture or cell-based methods
- sensitivity to surfactants, salts, pH, or media components
- downstream analytical performance
The relative importance of these factors depends on the target, sample type, biological requirements, and analytical endpoint.
Three Questions to Consider
1. What must be recovered?
Consider the target type, target state, sample characteristics, and whether the material is expected to be free, particle-associated, cellular, viral, or otherwise sensitive to recovery conditions.
2. What biological properties must be preserved?
If viability, infectivity, cell health, morphology, or functional state matters, recovery chemistry should support those requirements.
3. What happens after recovery?
Consider extraction, PCR, qPCR, digital PCR, sequencing, culture, imaging, flow cytometry, cell-based assays, or other downstream methods.
A formulation that works well for one analytical endpoint may not be the most useful starting point for another.
How the Standard Elution Fluids Differ
PBS
A broadly compatible biological buffer and a logical starting point when culture, viability, infectivity, or multiple biological endpoints are important.
Tris
A formulation oriented toward molecular workflows, including nucleic acid extraction, PCR, qPCR, digital PCR, sequencing, eDNA, metagenomics, and related applications.
PBS-X
A specialty Tween-free formulation for workflows where compatibility with sensitive biological targets or analytical methods is important. Examples include viral infectivity studies, extracellular vesicles, mammalian cells, imaging, flow cytometry, and other Tween-sensitive workflows.
DMEM
A specialty cell-culture medium for workflows involving mammalian cells, cell viability, functional assays, or direct downstream cell-based analysis.
One Formulation May Support Multiple Endpoints
Elution Fluid selection does not necessarily require a different formulation for every downstream test.
For example, a biological workflow may require preservation of viable target material while also using a portion of the recovered sample for molecular analysis. Appropriate nucleic acid extraction can allow several formulations to support PCR, digital PCR, sequencing, and related molecular methods.
Selection should therefore reflect the complete analytical plan, not only one downstream step.
When compatibility is uncertain, evaluate the proposed formulation with the intended downstream method before processing critical or irreplaceable samples.
When Workflow-Specific Evaluation Is Useful
Additional evaluation may be appropriate when:
- the target is particularly sensitive
- more than one biological endpoint must be preserved
- the downstream assay has known buffer or surfactant sensitivities
- the matrix introduces unusual chemistry
- the workflow combines culture and molecular analysis
- a specialized formulation may be needed
