Carbonated Beverage 4C Workflow Guidance for CP Select™ Sample Preparation

Carbonated beverages present unique sample preparation challenges for microbial detection workflows. Dissolved carbon dioxide, foam, yeast, protein haze, polyphenols, hops, colorants, fruit solids, sweeteners, and other matrix components can affect concentration performance, target recovery, and downstream assay compatibility.

These challenges are especially important when using rapid microbiological methods such as qPCR, dPCR, sequencing, flow cytometry, culture, enrichment, or other validated downstream workflows. Only a small portion of the original sample may ultimately reach the analytical method, so preparing and concentrating the sample can make a meaningful difference.

The Carbonated Beverage 4C Workflow Guidance document helps users evaluate beer, wine, cider, hard seltzers, RTD alcoholic beverages, soft drinks, sparkling waters, energy beverages, functional beverages, and other carbonated or fermented beverage matrices before concentration with CP Select™.

Why Use the 4C Workflow?

Successful concentration of microorganisms from carbonated beverages is rarely just a filtration step. Each beverage matrix behaves differently, and the best workflow depends on the product, target organism, concentration tip, elution fluid, and downstream method.

The 4C Workflow provides a structured way to evaluate the sample, prepare the matrix, reduce process interference when needed, and concentrate the target into a smaller, analysis-ready volume.

The goal is to balance four outcomes:

  • Target recovery
  • Sample processability
  • Concentration factor
  • Downstream compatibility

Characterize

Evaluate beverage composition, carbonation level, pH, alcohol content, haze, particulates, yeast or sediment, color, fruit solids, and expected microbial burden.

Characterization also includes defining the target and downstream method. A workflow designed for bacterial spoilage detection by qPCR may not require the same preparation as a culture-based workflow, sequencing workflow, or method focused on yeast or other larger targets.

Condition

Conditioning prepares the sample for concentration. For carbonated beverages, degassing is often the most important conditioning step. Excess dissolved gas can cause foam, unstable flow behavior, air entry into the concentration tip, and inconsistent processing.

The InnovaPrep® BeFlat™ Degassing Jar can be used to reduce dissolved CO₂ before concentration with CP Select™. Other validated degassing methods may also be used when appropriate.

Some beverage matrices may require additional conditioning. Examples include controlled pH adjustment for selected protein haze conditions, dilution for difficult matrices, or other matrix-specific preparation steps.

Clarify

Clarification may be useful when haze, yeast, particulates, fruit solids, or other matrix components prevent practical concentration or interfere with the downstream method.

Clarification should not be treated as a default requirement. Any clarification step may change where the target is located or reduce recovery if targets are associated with particles, yeast, sediment, or haze components.

Concentrate

After characterization, conditioning, and any needed clarification, the sample is concentrated using CP Select™ and Wet Foam Elution™ (WFE) recovery.

CP Select™ uses single-use Concentrating Pipette Tips to process beverage samples and recover retained targets into a smaller final volume suitable for downstream analysis.

The resulting concentrate may be used for:

  • Culture-based methods
  • Enrichment workflows
  • qPCR
  • dPCR
  • Sequencing
  • Flow cytometry
  • Other validated workflows
